Tumors that affect the endocrine functions of the pancreas are called neuroendocrine or islet cell tumors, but these are fairly uncommon. These tumors are named for the type of hormone-producing cell that is initially affected. For example: insulinomas (insulin), glucagonomas (glucagon), gastrinomas (gastrin), somatostatinomas (somatostatin), and  VIPomas (vasoactive intestinal peptide or VIP). Functioning islet cell tumors still make hormones, while non-functioning ones do not. Most of these tumors are benign, but non-functioning tumors are more likely to be malignant, islet cell carcinomas.

Man made (synthetic) corticosteriods are used to treat a large number of conditions and symptoms. Corticosteriods are used as a replacement therapy when the body is not naturally producing enough of its own of natural corticosteriods. They are also used to treat conditions where there is inflammation, autoimmune conditions or allergy symptoms. Corticosteriod can be taken orally as a systemic treatment to treat the body as a whole or it can be applied to the affected area for a local effect as creams, inhalations, nasal sprays, eye drops, ear drops or injections. Examples of conditions they treat are allergies, asthma, COPD, dermatitis, ulcerative colitis, Crohn’s disease, multiple sclerosis, nephrotic syndrome, adrenal insufficiency, inflammation, Addison’s disease, rheumatic conditions such as arthritis, fibromyalgia and lupus.

Failure of MC2R to activate in response to ACTH causes familial glucocorticoid deficiency (FGD), a rare autosomal recessive disorder characterized by severe cortisol deficiency with high plasma ACTH levels and normal mineralocorticoid levels (see "Causes and clinical manifestations of primary adrenal insufficiency in children", section on 'Familial glucocorticoid deficiency (FGD)' ). MC2R mutations resulting in effective loss of the receptor function are responsible for FGD type 1, which accounts for up to 25 percent of all FGD cases [ 4 ]. The melanocortin receptor accessory protein (MRAP) and its paralogue MRAP2 are small single-pass transmembrane proteins; MRAP is a key accessory factor for the functional expression of the MC2R /ACTH receptor. The clinical observation that 20 percent of FGD cases are due to inactivating MRAP mutations highlights the importance of MRAP in adrenal gland physiology [ 5 ].
